MACS Tissue Clearing Kit

The MACS method can obtain 3D neural structure information of the whole mouse brain and embryo by combining multiple fluorescence labeling methods and illumination imaging. At the same time, MACS method has good compatibility with lipid soluble membrane dye DiI, so it can realize the reconstruction of 3D vascular network of brain, spinal cord, spleen and other tissues and organs in normal mouse, and 3D visualization and quantitative statistical analysis of glomerular pathological structure in diabetic mouse.

Product presentation

The MACS in vitro clearing reagent developed by Jarvis can be used to transparent all kinds of in vitro tissues and organs such as heart, liver, spleen, lung, kidney, brain, tumor and organoid of scientific research animals. Combined with optical imaging, the 3D overall image results of all kinds of in vitro tissues and organs can be obtained. This in vitro clearing reagent has a fast transparent speed, such as only 2.5 days for the whole brain of transparent mouse; good transparent effect, the overall transparent effect is better than the common clearing reagent on the market; at the same time, it is compatible with all kinds of fluorescent dyes.

Directions for Use

Instructions for use of the MACS in vitro clearing reagent 

1. Composition appearance: This product is divided into three parts, R0, R1, R2 are colorless transparent liquid, R2 is colorless to light yellow slightly viscous transparent liquid.

2. Main components: R0, R1, R2 are mainly composed of MXDA, sorbitol, water and so on.

3. Storage: Keep away from light at room temperature, shelf life is 6 months, please use as soon as possible after opening the cover.

4. Operation instructions:

4.1 Sample soaking time: MACS-R0,1-1.5 days; MACS-R1,0.5 day; MACS-R2,0.5 day.

4.2 All the above three steps must be oscillated at a constant temperature of 25-30°C.

4.3 The sample should be placed in 5-15ml brown vial, and the amount of reagent should be twice as much as that of the original sample at each step. If the original sample needs 5ml, it is better to add 10ml transparent reagent (the average single dose of each sample is 4-10ml, and the specific amount depends on the sample size).

4.4 After fixation with 4% PFA, the samples should be thoroughly cleaned with PBS (>12h) and then transparent.

4.5 Sample should be immersed in MACS-R2 reagent when it is imaged.
